Size determination of unknown protein using the Ferguson plot
During electrophoresis in SDS-polyacrylamide gels there is a linear relationship between the logarithm of the molecular weight and the relative distance of the migration of the SDS-polypeptide micelle.
To calculate the molecular weight of an unknown sample or to identify the correct protein of interest, a Ferguson plot can be used. The relative distance of migration (Rf) value of the markers are plotted against the log molecular weight (Mr) to create a standard curve. From the standard curve and the Rf value of the sample(s), the relative molecular weight can be calculated.
Note: Size determination can easily be performed automatically using ImageQuant™ TL software.
The Rf values are calculated from the
wells (Rf = 0) down to the dye front (Rf = 1).
