ACID PICRO MALLORY

1. Bring sections to water

2. Stain in Celestine blue, 3 mins

3. Wash in water

4. Stain in Carazzi Haemalum 3 minutes 5. Wash in water

6. Blue in Scotts tap water

7. Wash in water

8. Rinse in 95% alcohol

9. Stain in Picro-orange 2 mins

10. Wash in water until only red blood cells are yellow

11. Stain in acid fuchsin 5 mins

12. Wash in water

13. Stain in equal parts Picro-orange and 80% alcohol

14. Differentiate in phosphomolybdic acid for 8 mins

15. Wash in water

16. Stain in analine blue, 8 mins

17. Wash in water

18. Dehydrate, clear . Mount sections in DPX


RESULTS:

--Fibrin - Bright red

--Muscle - Red

--Collagen - Blue

--Red blood cells - Yellow/Orange


SOLUTIONS:

1. 0.2% orange G in saturated alcoholic Picric Acid

2. 1% Acid fuchsin in 3% Trichloroacetic Acid

3. 1% Phosphomolybdic Acid

4. 2% Analine blue W5 in 2% Acetic Acid (soluble Blue)
