Rapid Coomassie Staining Procedure

  1. Make up following solutions (maybe stored for months at room temp.) Ratios are v/v unless stated otherwise.
  2. Stain:

    40% methanol; 10% acetic acid; 50% water; 0.1 % (w/v) Coomassie Brilliant Blue R250

    De-stain:

    40% methanol; 10% acetic acid; 50% water; 2 Kim Wipes



  3. Begin staining (Procedure is for mini-gel; takes about 10 minutes to see results; use longer incubation times for thicker gels)
  1. Put your gel in a plastic staining container with lid
  2. Add enough Stain to cover gel well
  3. Put lid on, microwave on high for 1 minute
  4. Stain while rocking (up to ten minutes, strong staining is usually seen after 4 minutes; 2 minutes is good to just detect proteins)
  5. Pour out stain, add a large amount of de-stain, add Kim Wipes.
  6. De-stain on rocker at room temp.
  7. Should be able to see results in a few minutes after adding de-stain; leave 2 hours to overnight for complete de-staining
