help for bacteria endospores fluorescent stain - (Sep/24/2005 )

Hi all, I need to see a bacteria endospore under fluorescent microscopes. However, the fluorescent dyes used for this purpose is kind of difficult to find. Any suggestions that which dye I should use? Also, to increase the binding between the fluorescent dye and endospore surface, any pre-treatment I should do before adding the dye in?

As endospores are so tough and have a relatively impermeable cell wall, getting a dye in may be difficult

Your easiest approach may be to use a general fluorescent stain eg. FDA, DAPI - anything that will stain the interior of the cell - but leave the spore dark - at least then you can see its outline!

This approach is what's used in ordinary endospore stains.
