transient transefection - plasmid dilution (May/12/2005 )

During transfection (transient),what happens to the plasmid inside the cells.During cell division do they get diluted?
Why is it said like treansfection is done with 70-80% confluence?Is it like more particles are taken up by more cells so u have a high transfection efficiency?

during transfection, assuming you are starting from a 70-80% confluency, plasmids in the cells aren't that diluted. In most cases transfection is quite good and allow to see results. If you are in trouble with that, you can transfect cells and pick 1/4 to make a selection. You will be able to see if differnces occurs and evaluate your transient transfection.

70-80% confluency is the good ratio between plasmid numbers and cell number.

Hi,

Plasmids do not get diluted in cell division, assuming that they include an "Ori" or Origin of Replication. This enables the replication and propagation of the plasmid as you cells grow and divide. I.e. as your culture grows, your cells duplicate the plasmid so that the progeny of any transfected cell will also contain the plasmid.

You transfect at 70-80% confluency because this is when the cells will be actively growing (in log phase) and transfection will be most efficient.
