What is overexpression, I know but my concept is not so clear. We increse the expression of a gene by overexpression and than compare it with wild type. But how could we know where the TDNA has been inserted? And the gene that we insert is expressed along with the indigenous gene to make more proteins? I mean if some could refer some paper or explain in simpler terms, I'd be greatly thankful .
I'm at a place where language is a big barrier for me to discuss with others, so if you find some foolish questions in future than please don't mind.

there are many indirect ways you can check that the insertion has been made, e.g. northern blot, gfp, and many others. whenever in doubt, you should go here first of all.
