May I ask what's the toxicology of the nicotine to microorganisms?
And,can microorganisms tolerate higher concentration of nicotine in solid substrates(e.g. tobacco straw, tobacco leaf) than in the agar medium?

Excessive smoking of the scientist does not stop fungi from growing we tested in my boss' office
Serious now: I would say it depends on the species. Alkaloids in general can interfere in membran stability and transport and/or interact with cellular processes of microorganisms, but this depends on the species. For sure it has not the same mode of action in microbes than in humans (no neurons in microbes)!
So always remember: mircrobes are capable of living and suviving in nearly every habitat and all environmential conditions and can do nearly everything (besides watching TV probably, but we do not for sure
If you are interested in a special species or group of microbes you should just try if and how much they tolerate
