the curious case of the disappearing x-ray bands - (Aug/28/2008 )

Hi! so something very weird happened to my western blot x-ray films. i developed them a couple of weeks ago and i looked at them today and all the bands had faded away:( does anybody know what happened?!! the blots were stored in my drawer, if that helps. i would appreciate any suggestions smile.gif

perhaps it got rubbed of? The layer of photosensitive emulsion can be rubbed off the plastic.

Drying membranes is never a good idea, usually causes high background and low signal. Store membranes in block solution in a small plastic pouch at 4 deg C.

Perhaps your fixative solution was not good anymore? Never happened to me. And I always store my membranes dryed (PVDF), no problems with background/signal ratio.

if the film is not properly fixed after development then it will fade more rapidly than normal (photographic emulsions fade over long periods of time).
