Yield after column purification - (Aug/06/2008 )

Hi all,

Has anyone else found that after column purification the spec gives very low to non existent readings yet the purified DNA band on a gel is visible and strong?

The reason given to this particular labmate who had this problem yesterday was that the purification column can leave particles in the DNA which will interfere with the spec reading and that centrifugation prior to checking the conc. will help.

I personally havent heard this before and have successfully been using columns for purification for years!


Also, i cant find anywhere in the literature that suggests this is true so I thought I'd put it out there to all of you folks biggrin.gif

YES that happen with me. it gives me OD zero but appear very clear band on the gel.

nope, I have never seen this happen before. This is the first time i have heard somebody make this particular report.
