PC12 cells are not "round up" - (Jun/02/2008 )
Hi, all,
Our lab recently purchased PC12 cells from ATCC and I cultured them with DMEM with 10% FBS and 5% HS, on CellBind plate, regulate tissue culture plate and poly-l-lysine coated plate. The problem is, my PC12 cells grow like slander "fibroblast" cells: they seemed to attach fine on any of the plates, they grew pretty fast (definitely divided more than once per 3 days), and only less than 20% of them "round up" like other paper said. I don't know what did I do wrong here. I tried to differentiate them using 5, 100, 250, and 500ng/ml of NGF with the same growth medium (well, I just learned that I should use low-serum medium) and only less than 10% of cells extended "neurite" like processes.
Basically I pretty trust cells from ATCC, so I think I must have done something wrong?

Thank you very much, that is a great website. However, I have checked them and I didn't find anybody talking about their initial culture being NOT "round" cells. Does anybody have anyway to "enrich" those round cells or other suggestions?
