Ca++ estimation in the cell

Ca++ estimation in the cell - (May/22/2008 )

I want to estimate [Ca++] in breast cancer MCF-7 cell line. Can anybody give me detailed protocol using Fluo-3 and Fura-2 from molecular probes ? I would like to know the precautions to be taken while estimating the [Ca++]
