Opinion about fluorescent antibodies - (Mar/18/2008 )

Hi everyone,

I've been searching secondary antibodies for use in immunofluorescence and I came across two new types of fluorochromes:

does anyone have experience with the new DyLights from Rockland or NorthernLights from RnD systems?

Another thing: which are, in your opinion, the best fluorescence conjugated antibodies in the market?

we use Alexa conjugated antibodies. They are good.

alexa secondaries from molecular probes are good.

We use the alexa secondaries that are highly cross-absorbed from molecular probes. A few more dollars for the extra purification but I have great success with these. Nice clean staining with minimal background.

We use Jackson ImmunoResearch secondaries raised in donkey - work great and no cross-reactivity.

I use Alexa and I've had mixed success. It's been hit and miss, I found some AMAZING ones with no background and no non-specific binding, but there were a couple that had terrible non-specific binding so no matter what run your controls.

I have also used jackson raised in donkey and had no problems, they just aren't as bright as alexa.

I am curious about trying dylights and since i do tons of IF I'll probably try them next
