anyone culture SKOV3 cells? what's the morphology under microscope? - (Mar/11/2008 )
Hi, anyone culture SKOV3 cells in vivo?
what should they look like under the microscope? should they look bright and round or dark star-shaped?
i culture some skov3 from frozen stock yesterday but they don't seem happy today and mostly are dead, don't attach to the bottom of flask.

Mine look star-shaped. I just split them today but I can give more detail tomorrow if you need. You may have either bad medium or a bad stock. What medium are you using?

The media I used was RPMI 1640 HEPES modification from sigma, supplemented with glutamin, P/S and FCS.
I took another SKOV3 stock today and cultured. After a few hours, there're some cells looked star-shaped and some were bright and round-shaped. But I'm not sure which ones were dead or alive. I cultured LoVo cells before, and they looked really bright and sphere under the microscope. Guess there must be variant morphology between these 2 cell lines

Sounds about right. The star shaped are cells that are alive and attached; the round shaped ones may simply be cells that are still attaching. Overall it sounds as if you are OK now and probably just had a bad stock the first time.
