I prepared a smear of bacterial colony in glass slide, then heatfix the bacteria.Can somebody tell me ,how heat fix the organism into the slide. What is the change that occurs in the structure of bacteria?Also, how it enables the stain to properly bind to the organisms?
Are you doing simple staining or gram staining? If your questions is how to heat fix the bacteria to the slide, then that is pretty simple. I usually attach a clothespin to the edge of the slide and pass the slide over the flame of a Bunsen burner about 3-4 times. When you pass the slide over the flame, pass it fairly quickly because if you leave the slide over the flame it will kill your bacteria. Off the top of my head I am not sure if there is any change in the structure of the bacteria due to heat fixation.
Thanks for the answer, but I mean to say what causes the microorganisms to adhere in the slide
Hmm...I'm not quite sure, but I thought it was simply that passing through the flame simply dries the bacteria to the slide. There shouldn't be a significant change in the bacteria, as that would affect what you see under the scope.
