in vitro model of epilepsy - cell culture (Nov/14/2007 )
Hi.
I am looking for an epilepsy model in hippocampal cell culture. it seems nobody has used glutamate or kainic acid on these cells. I just want to share my problem hoping somebody can help me.

Many people have tried experimenting by adding glutamate to the hippocampal neurons, it the classical neuronal death assay.
Well, I am not familiar that its being used for epilepsy.

We use KA in hippocampus slices 6 uM.

I really need your help. Actually we are looking for a model (thorough applying glutamate or kainic acid or any drug that can cause epilepsy) to induce cell death as a model of epilepsy. As these two can cause epilepsy.
But I did not find much paper regarding applying these two drugs to induce hippocampal cell death and then I'm a bit worried about the possibility of using them.
If you are applying these two and analysing cell death too, would you mind to explain more about the dose and the condition of drug exposure.

One has to make up a buffer to add to the cells. To the buffer one will add glutamate and glycine. The media from hippocampus is removed (and stored)and the buffer is added for a certain period of time and then replacement with the original media. You have to make different concentrations and analyze cell death and choose concentrations which suit your experiment.
I am not sure about the concentrations to use. But glutamate toxicity is a common thing and you should be able to find lots of papers.

Another question as I guess you are working with hippocampal cell. Do you think can we transfect these cells with an special protein?

What do you mean by 'transfect with a special protein'? I am guessing a transfection reagent. Its a bit tricky for neurons. But there are transfection reagents or electroporation devices (amaxa) for hippocampal neurons. But we have always used viruses as they give a very high transfection rate.

We only use viruses for tranfecting neurons. So I donot have protocols with transfection reagents.
