RNA concentration issue for miRNA array - (Oct/21/2007 )

Hi everyone,

Is it posssible to concentrate RNA for miRNA microarray? My sample has a RNA concentration of about 150ng/ul and I need a concentration of 1ug/ul for miRNA microarray! Is it possible to concentrate my sample?

Thanks in advance for your replies...

Thanks,

You could try the Qiagen RNA Elute kit, it worked fine for me. You put your RNA on a coloumn again in a larger volume and elute it in the end very clean and concentrated in only 14 µl.

You could also try EtOH precipitation - Ambion has a protocol - http://ambion.com/techlib/misc/acetate_precip.pdf (this is what I used - but be careful - if you use the flashPAGE, there is a LOT of salt in the buffer leading to a massive pellet that should be washed several times).

Qiagen RNA Elute kit is good for concentrating your sample.
I concentrated many samples by this kit and it worked for me.

I use a concentrator like this

works quick and easy, less losses than with column based kits,
