transfection reagent for HUVECs ? - high cytotoxicity problem (Oct/13/2007 )
Hello friends
Please suggest me to find the best way to trasfect plasmid DNA to primary endothelial cells, HUVECs. I have tried some of the commercially avaiable reagents but failed due to high cytotoxicity. I used Genejammer, Lipofectamine, and Effectine previously for the same.
I will be thankful to get your valuable suggestions and name of reagents from your experiences.

i suppose you had tried at different concentrations of DNA and lipids. well there is also Roche's Fugene (worked well for the HUVECs in my lab) but lipofectamine also worked well in my lab. Did you try the L2K or the LTX? LTX gave higher readings for me though.
L2K works on this type of cells with ~30% transfection rate with a some toxicity. The expression appears to be transient with a rapid decline after 36 hr. LTX should be less toxic according to Invitrogen, although I have not tried it.
