Longest single DNA sequencing read? - (Sep/19/2007 )

I am wondering if anyone knows what the record is for the longest single DNA sequencing read? The reason I ask is I have a trace that gave me over 1400 Q20+ bases and I am wondering if I have the world record rolleyes.gif

mmmm. well my standard reads are rouhly 400-500 bp, but using a company, i got nice 900bp readings...
how damn did you succed this record ?

Thanks for noticing I have been missing smile.gif I have been busy working on a new DNA basecaller called PeakTrace. This basecaller is how I managed to get such a long read. I am in the process of writing up a paper on it and it occurred to me that 1400+ bases is probably the record, but I can't remember what the previous record was (I have a vague memory that someone claimed 1100+ bases in a paper I read a few years ago). If anyone can remember what this paper was then I would be very grateful.

that beats the record for my lab.
The record here was 1200+ bp.

You haven't published this traces by any chance? On what instrument was it generated?

No, I have not published that group of traces. And sadly I am not sure what machine was used either, although I do know it was a machine at Oxford sequencing.

Prior to Oxford University Sequencing's privatisation, my lab used to send it Big Dye preparations to be run there. After said reorganisation the fantastic long reads stopped, the prices when up and my lab had to go looking for somewhere else. Never did get those long reads again. 1100bp was the best I have seen since the move.

Such is the way with sequencing. Although it is not as bad a the 377 days sequencing quality is very dependent on the person performing the run.

If you are interested in getting the maximum quality have a look through the reviews of DNA sequencing service providers we have on our site. I can't really recommend who to use in a public forum, but if you want to PM me I can give you a couple of recommendations. Anyway the link to the reviews is below.

We have 700-900 pb more or less, ones in a while, 1000 pb, no more
