T antigen status of human cells for transfection - (Aug/07/2007 )
Sorry i have a very basic question that needs answering and was wondering if anyone couldhelp me out with it.
I want to transfect human synoviocytes with an protein expression plasmid containing a T7 promoter.
Is it likely this plasmid would work in this cell type?

What T antigen? SV40 T antigen or T7 phage RNA polymerase? Primary cells do not have either.
Is the T7 promoter only promoter it has in your plasmid? You have to either have an eukaroytic promoter, such as CMV promtor to work, or if you can supply T7 polymerase through another expression mechanism, providing that it also have an internal ribosome entry site 5' to your coding sequence, to express this sequence from this plasmid.
