DsRED fluorescent protein - (Jul/18/2007 )

Hi Has any one worked with dsRED fluorescent protein in mouse embryos?I injected the DsRED mRNA into mouse embryos and I didnt get fluorescence even though I used the right wavelengths.I used the pDSRED- Monomer C1 to make my mRNA. Please help me.Thank you.

Have you tried another reporter, luciferase mRNA, for example, or injection of expression plasmid DNA for desRed?

I've used DsRed in cell culture and got really low fluorescence also, apparently it's typical for DsRed that is not so fluorescent, as an alternative, we have used HcRed, which is a little better

I used EGFP and got high fluorescence. For my experiment I have to use mRNA which gives red fluorescence.

What is HcRED and from which company did you buy it?Compared to Green fluorescence do you know how much HcRED fluorescence will be?

dsred monomer gives a very low fluorescence. its takes time for fluorescence to come up and also very faint. We wanted to use it but because of these problems we change fluorescent proteins.

Dsred2 is better but can also be toxic.

Does your protein need to be monomeric and orange/red fluorescent? Or can you choose any fluorescent protein?
DsRed-monomer is one of the weakest fluorescent proteins around, so you better change your reporter to EGFP, AcGFP, or some of the ones created at Tsiens lab (www.tsienlab.ucsd.edu, Clontech now also sells some of these, but tsien has them cheaper for non-profit-labs if I'm correct).

Tseins lab gives them for free. But because of huge demand its could take a few months. Its better if you knew someone who has already received them and get it from them and send tsiens lab a MTA.
