Hi guys,

I guess I may have to do a triple transfection with three plasmids for HEK293T cells. I have done transfection before with only one type of plasmids, using lipofectamine. All the three plasmids have been successfully transfected to the HEK cell, individually or in pair, but never all together. I have heard some difficulties with triple transfection.

Could you guys clarify what kind of efficiency I should expect and how triple may be significantly harder than or different from double transfection? Thank you in advance.

Its not much of a problem. I regulary do quadruple efficiency for virus preps and dont have any problem. I have the total amounts of DNA same, so I would divide the total DNA by 3 for triple transfections.
