G418 resistant 293 - (Feb/23/2007 )

Are any of the 293 cell lines(HEK 293, 293 T or any other) resistant to G418?

Not normally. But 293 FT's are resistant to G418.

I've seen many posts in this website where people are discussing about conc of G418 for selection of transfected 293 cells which means that this cell line can be used for making stable cell lines.
But one of my professors said that all 293 cells are resisitant to G418 & u can't use these for making stable cell lines.
Im just confused abt this.

the normal HEK 293 are not G418 resistant. Some modified versions like 293FT are G418 resistant. We have made stables cell lines using 293's in our lab. How would you explain that?

May be the cells in your lab, could somehow be G418 resistant due to prior manipulations. Only the professor would know that. But if you could get a fresh 293 from anywhere else, would be useful for you.

I doing a killing curve for G418 disulfate salt, (((( I wonder what's the difference on just G418 or g418 sulfate,, I really dont know which one to use for stable cell lines, but I bought the G418, disulfate salt from Fisher, any suggestions?? ))))

The point is that my cells are not dying even with 900ug/mL, and they've been sitting for 4 days, I've been replacing the media every 2 days and nothing, they are just fine. I don't know if its that I have to many cells, ( I plated them in a 12 well plate, and they are pretty much confluent, like 90%)

What am I doing wrong?? Any suggestions in how to make a good killing curve??
I am using SKBr3 cells, breast cancer cell line.
