I have a very simple question but we could not agree on sth with my colleague.
Do you use PBS or Water to dilute Triton X-100 to have 10% Triton X stock which is to be used for the preparation of other buffers.

I dilute in water, so I can use it after in PBS solution or tris solution...

for a stock dilution of triton x-100 take distilled water; Na+, K+, Cl- or PO4- ions may disturb in other buffers
For better accuracy in preparing 10% Triton X-100 (in water), use the density (100% = 1.06 g/mL) rather than trying to measire volume with a pipettor.
Weigh 1.06 g of 100% Triton X-100 in a 15 mL Falcon tube.
Add water to a total of 10 grams. Mix.
Roche sells packs of 5 x 10 mL vials of very high quality 10% Triton X-100 specially purified to eliminate peroxides formed by oxidation.
we make up a stock sol. of 10% triton. Its better to pipette it this way.
