HPLC - Which column (Nov/30/2006 )
hello,
my college is planning to get hplc and i would liek to know which type of column is good for plant secondary metabolities and what size of the column gives better resolution of the metabolities.
the hplc will be mainly used to purify the secondary metabolities from extracts that show antimicrobial activity against the tested organisms. as there are so many different columns available with various sizes, i would like to knwo the type of column and the size to be used for this kind of work.
any suggestion or any websites or books giving information about this would be great,

To select a column you need to know the Mr and physical properties of the molecules you want to analyse. e.g. what's the solubility, polarity, hydrophobicity, pKa and so on. You'll learn so much about your molecules going through this exercise.
Best thing: pop them into a science search engine and add HPLC. In the method section of the papers you'll see what other researchers have used.
Alternatively, contact a supplier for their catalogue. They usually have a section on column selection.
