Protocol for reuse of PD-10 desalting columns - (Oct/09/2006 )
Hi,
I'm looking for a wash protocol to reuse prepacked PD-10 desalting columns from Amersham. I use them to desalt protein extracts from plants.
Has somebody a good (and easy) protocol to clean the column afterwards for reuse?

As good as I know those columns are filled with Sephadex G-25 so you don't really need any special protocol. It would be enough to just wash them with few column volumes of your buffer (or even water). You can use conductometer if you really want to check it you have washed out all salt. To remove tightly bond impurities use two column volumes of 0.2M NaOH. If you need to store your columns add 0.02% of sodium azide to your buffer.
