Positive Control for apoptosis - (Aug/07/2006 )
Hi everyone,
I just started my study on effect of virus to the glioblastoma cell line (U87MG). Literature said the virus able to inhibit proliferation of the tumor and most probably the mechanism is apoptosis. So, anyone can suggest me a positive control for in vitro and in vivo? Thank you very much!
With positive control you mean something that will surely induce apoptosis...right? We use Staurosporine with the cell line U87MG.
U need to induce apoptosis in ur cell line and also compare it to the virus infected ones to check for cell death.
U will have to show the presence of caspase activity in the cell line to show that the cells r dying due to apoptosis. people use different chemicals to induce cell death invitro, eg. glutamate, low glucose or hydrogenperoxide etc.
how do u want to study invivo? R u planning to inject the tumour cells in to the brain or do u have a mice model for the tumour? If so then inject the same chemical into the tumour or any other thing drug used for chemotherapy -administer systemically. It depends on how u have planned the experiment.

Yes i need something that surely can induce apoptosis in vitro and in vivo. I will induce the tumor in nude mice. Thanks Jou, I will look up on Staurosporin.
Erm...Scolix, u mean if we use glutamate for in vitro (example MTT assay), we also can use glutamate to be injected in nude mice? Any suggestion for reference? Thank you ! ![]()

U could use glutamate for invitro (no doubts abt it), and u could check its usage for invivo as well.
