Separate glycoproteins and glycosaminoglycans - (May/16/2006 )

Hi everyone!

Recently, I did an experiment of extracting glycosaminoglycan from urine. However, I didn't know how to separate the glycoproteins with glycosaminoglycans. This impeded the migration of bands in agarose gel electrophoresis.

I am thinking of using lectin which binds carbohydrates to separate glycoproteins and glycosaminoglycans, but which lectin should I use if I don't know the glycoproteins contents? Please teach me how to get rid of those glycoproteins.

a good starting lectin would be concanavalin A (con A). it binds mannose and glucose (and galactose, i think). many glycoproteins contain mannose residues. don't forget to add Mn and Ca to the buffer.
