Apoptosis inducing reagents - (Apr/04/2006 )

Hi everybody,
I would like to find out some chemicals that are commonly used as the positive control for apoptotic induction in K562 cells.
Could anyone help me? Is Ara-C ok? And which supplier is available?

How about staurosporine?

Besides, as I know, etoposide is an antiproliferation drug. I wonder if this drug can induce apoptosis?

hi,

etoposide-treated cells appear positively stained for annexin V and negative for DiOC(6)
but i don't know the effect of staurosporine on K562 cells... i can't help you more wink.gif

I'm not very familiar with AraC, although staurosporine is a general tyrosine kinase inhibitor. While it does cause apoptosis, the mechanism isn't well defined and I would generally avoid it. If apoptosis alone is the endpoint you are measuring you may be interested in using bleomycin, etoposide (as mentioned), 5-fluorouracil, UV or gamma irradiation or a host of other compounds.
