DNA purity for ligation - (Mar/26/2006 )
my vector purity is 1.9 and my insert purity is 1.2
i ligate these two and then perform transformation , mini prep and large prep
from this large prep sample i isolate a fragment containing the insert fragment of purity 1.3
is there will be any problem in further ligation with this isolated fragment???

basically 2ligations are 2 diffferent things. But as your first was succesful, i assume the other one will be too.
