How to design Rescue control - (Feb/27/2006 )

Hi,
Today I was asked to design a RNAi rescue control. However I have totally no idea which sites are essential in my siRNA. And at least how many sites I should mutant to block siRNA binding. So, does anyone have experience in such an experiment. Thank you in advance.
