how to confirm if ligation is sucessful? - (Dec/13/2005 )

Besides transforming the ligation products into competent cells and select, what other ways that I can use to confirm ligation is successful?

Well I guess this is the only real test, some peoples run the ligation product on an agarose gel where you can see the ligated products compared to the single bands of individual vector and insert but this doesn't indicate that you will generate recombinants but rather that the ligase has done her job (at least to a certain extent)

You can do quantiative PCR with a pair of primers priming across the ligation gap. We are doing this to track cutting and religation of plasmid assemblies.

i run on agarose gel the liation and the control without insert. Presence of different profiles MAY suggest a working ligation.

After Transformation instead of isolating plasmid and digesting and checking it you can do a clolony PCR.
