DNA extraction from saliva/buccal swabs - (Apr/28/2009 )
Has anybody on here any experience with and/or OK protocols for DNA extraction from buccal swabs or saliva, either manual extraction or automated with extractors like M48, magNA Pure etc? Any help would be appreciated.
What is a normal yield from such DNA extraction?

I suggest you look into our AquaGenomic protocol. If extracting from mouthwash, you will get tons of DNA.
I actually found that using simple phenol chloroform extraction worked the best after treating with proteinase K to killany proteins. It depends how much buccal swab product there is, but if there is a fair bit, then i acheived yields of ~500ng/ul+
Phenol-chloroform produce dangerous wastes, I prefer kits that reduce this problems.
