RNA extraction using Sigma lysis buffer and Qiagen spin columns - (Nov/20/2018 )
Hi all,
I was wondering if anyone has tried using interchangeably the Sigma lysis buffer from Sigma’s GenElute Mammalian Total RNA Miniprep Kit and Qiagen's AllPrep DNA/RNA Mini Kit.
I lysed my organoids in the Sigma lysis buffer instead of RLT, but was wondering whether I could still use the Qiagen columns in order to extract DNA..
Anyone tried this?

I hope you will hear from someone who has actually tried this. I have not, and I generally advise against using components from different branded kits in the same prep. However, the two lysis buffers both contain salts of guanidine (complete compositions are proprietary) and you add beta-mercaptoethanol to both, so they are similar. Since you have already done the lysis, I’d try processing just 1 or 2 (for balance) through the Qiagen system and see what you get. If it doesn’t work, and the rest of the samples are hard to replace, get the kit that matches the lysis buffer to finish them. Maybe you can borrow some from another lab? Good luck!
