Methylene Blue Solution

Methylene Blue Solution (Loeffler's) is used in various procedures such as the Ziehl-Neelsen, Fite's, and Gram stain. The stain is also suggested for use in staining a variety of bacteria (Corynebacterium diphtheria, Haemophilus influenzae, Neisseria, etc.) and leukocytes. This reagent produces dark blue nuclei with lighter blue cytoplasm.

Nuclei: Dark Blue
Cytoplasm: Light Blue
Corynebacterium Diphtheria: Blue Granules
Escherichia Coli: Blue Rods
Streptococcus Pyogenes: Blue Cocci

Urinary sediment supravital stain

In my work, used a 2% methylene blue solution or 0.4% toluidine blue (from 1% stock solution dilute in dH2O) as a simple quick supravital stain, using as support a microscope glass, dispensing on a sediment drop, one drop of these stains and mix helping by plastic stick employed to mix coffee drink.

Solution I : dissolve 3.0 g crystal violet in 20.0 ml 95% ethyl alcohol and the dissolve 0.8 g ammonium oxalate. Stable indefinitely at room temperature.

Solution II: dissolve 1.0 g safranin O in 40 ml 95% ethyl alcohol and the add 400 ml dH2O. Stable indefinitely at room temperature

Stain work solution: mix 3 ml of solution I with 97 ml of solution II and filtered. The mixture showld clarified by filtering every 2 weeks and discarded after 3 months.

Procedure

Add one or two drop of stain to approximately 1 ml of concentrated urine sediment. Mix with a pipet, and place a drop of the suspension on a slide and coverslip.
