I'm new to site directed mutagenesis and would appreciate all the help. 

 

I need to perform a single gene mutation. The antibody against my protein target is Ser552. I want to change the Ser552 to another amino acid, I assume Ser552 means serine at position 552 from the first amino acid. But the antibody data sheet states the serine at a much different position (i.e. 853), much further down the sequence. am'I missing something?

Are there splice variants for this gene? Or it could be that you are looking at a different species sequence, or the supplied sequence is wrong (mis-translated or starting from a different ATG).
