How to establish drug-resistant cell lines? - (Jul/08/2015 )

Greetings!

I was wondering if any of you has worked on establishment of drug resistant cell lines? I want to know how to generate drug resistant cell lines? Can anyone provide me the protocol for it? I need to establish doxorubicin and cisplatin resistant human lung cancer cell line (NCI-H460). Help! Thank you.

You could do this by culturing under increasing amounts of the respective drugs (probably one at a time), thereby selecting for cells that survive the conditions.  Another method would be to insert the resistance genes required by conventional methods such as recombination or CRISPR/Cas and then selecting for the inserted genes.

 

In both cases you need to ensure that the cells you have at the end are the same behaviorally and physically as the parent line. Also, don't expect this to be quick. I would be looking at a year or more for both instances.
