Weird gel banding for apoptosis - (Jun/30/2013 )

Hey guys,

I'm currently doing an apoptosis assay using the protocol from http://www.celldeath.de/apometh/dnaladd.html.

And when run on gel, the results I got had some smearing above the well. Anyone has encountered this before and know why did this happened?


L1: 1kb ladder
L2: Negative control (no apoptosis)
L3: Positive control (apoptosis present)

Your DNA has stopped in wells because Proteinase K has not done its job completely.

Do you have any idea about the present of smearing above the well too?

I don't think that what you see above the well is smearing, but just the Ethidum bromide running in the opposite direction to the DNA. (Assuming you are using EtBr).
