How can we determine the allosteric regulation of an enzyme experimentally?

the short answer is: assay the enzyme

longer: if positive substrate cooperativity, then substrate saturation kinetics will show a sigmoidal curve (as opposed to hyperbolic)

if cofactor then assay with and without said cofactor

if negative cooperativity, then inhibition assays
