Best way to do Immunohistochemistry - (Nov/28/2012 )

Im new working in histology...im trying to do immunohistochemistry or LacZ staining but dont have good results...

i like to know the best ways to fix, dehydrated, stain, cut, etc...

These all depend on what exactly you are trying to do and what tissue(s) you are using...

im using mostly spleen and lymph nodes (mice) and i want to know the presence or location of a protein of interest... so i need good cuts to analyze my protein, but i have mostly "broken or fragmented" tissues...
