IHC nuclear staining - hematoxylin (Aug/11/2010 )

I'm new to IHC staining and I woul dlike to check the localization of b-catenin (whether its nuclear or cytoplasmic or both). I'm doing IHC staiing with the antibody folowed by hemotoxylin staining? Is this correct or do I have to do H&E staining to see the cytoplasmic and nuclear staining both?

You should do both, otherwise you will be unable to determine the extent of the cytoplasm, in case you find it in the cytoplasm as well as the nucleus.

Are you going to try an antigen retrieval step for the nuclear staining? I had to do this for the antibody I produced - so just a warning ;)
